Sunifiram is incredibly concentrated compared even to the strongest racetams like Pramiracetam. You will see positive effects from a dosage as small as 4 mg though many people typically take between 5-8 mg at a time. A Sunifiram dose of 10 mg is more likely to cause a stimulant effect similar to Adderall. Taking a 10 mg Sunifiram dosing is not toxic and not likely to cause serious negative side effects, however you may experience increased body temperature and perspiration. - Mechanisms of action of sunifiram
How does sunifiram work? Preliminary studies suggest Sunifiram has two main mechanisms of action and in the below four steps:
- Sunifiram activates AMPA-mediated neurotransmission.
- It enhances LTP in a bell-shaped dose–response relationship. This enhancement by sunifiram is associated with an increase in phosphorylation of AMPAR through activation of protein kinase II (CaMKII) and an increase in phosphorylation of NMDAR through activation of protein kinase C α(PKCα). More specifically, sunifiram stimulates the glycine-binding site of NMDAR with concomitant PKCα activation through Src kinase. Enhancement of PKCα activity triggers hippocampal LTP through CaMKII activation.
- Sunifiram improves cognitive deficits via CaM kinase II and protein kinase C activation.PKC activation may be a common mechanism amongst cognition stimulating drugs from different chemical classes.
- Sunifiram aids in the release of acetylcholine in the cerebral cortex.

Currently, there is not any known toxicity that is associated with Sunifiram. Scientific research has also determined that animal test subjects have a tendency to build up a tolerance to Sunifiram on a rapid basis. This process of tolerance is slowed down when Sunifiram was introduced sparingly as opposed to a consistent basis.
Additionally, scientific research on animal test subjects have yielded a minimal amount of side effects in relation to Sunifiram’s usage, and it has been theorized that the side effects that do exist relate to pain that is consistent with headaches.
